Flavonoids distributes in the nature extensively, mainly is exist in the leaf and fruit of theplant. Flavonoids have much beneficial living activity which urged people's research toflavonoids into a new stage. Lonicera edulis is a kind of natural, green, wild edible berry.Lonicera edulis have the function of anti-inflammatory, decline blood pressure, antioxidant andanticancer etc. According to the report, the main activated composition of Lonicera edulis isflavonoids (anthocyanin). This thesis mainly studied flavonoids in Lonicera edulis onextraction, purification and effect test.This thesis adopted three different methods to extract flavonoids from Lonicera eduIis.The optimal process and optimal process were ascertained by comparing and analysis.The optimal process on extractive method with water was ascertained by the single factorexperiment and the orthogonal design. The optimal extraction conditions were that theextractive temperature was 70℃, the extractive time was 6h and the ratio of materiel to liquidwas 1:100. The optimal process on extractive method by ultrasonic was ascertained by thesingle factor experiment and the orthogonal design. The optimal extraction conditions werethat the ultrasonic response time was 7min, the ultrasonic power was 70℃, and the ratio ofmateriel to liquid was 1:100.The optimal process on extractive method with ethanol was ascertained the single factorexperiment and the orthogonal design. The optimal extraction conditions were that theextractive time was 7h, the extractive temperature was 60℃and the concentration of ethanolwas 50%(v/v).The response ichnography design was chose which was used to the farther study of theoptimal process on extractive method with ethanol. The optimal extraction conditions were thatthe extractive time was 6.6h, the extractive temperature was 61.3℃and the concentration ofethanol was 55.3%(v/v). In these conditions, the extractive weight of flavonoids was64.94mg/g. The conclusions were that the optimal extraction method was extractive methodwith ethanol and the most extractive weight of flavonoids was 64.94mg/g in the optimalconditions.The Lonicera eduIis flavonoids were purified by AB-8 macroporous adsorption resin inthis thesis. Some factors such as extract sample pH value, extract sample velocity, extractsample concentration; eluting ethanol velocity and eluting ethanol concentration were studied.The optimal purification conditions were as following: extract sample pH value 4~5, extractsample velocity 60mL/h, extract sample concentration 120~150mg/mL, eluting ethanol.velocity L 60mL/h and eluting ethanol concentration 40~50%(v/v). The purity of the Lonicera eduIis flavonoids product reached more than 65%in the technological conditions.The effect on reducing blood lipid of Lonicera edulis flavonoids possess was confirmedthrough fed Lonicera edulis flavonoids to the rats and determinedthe weight of the rat andblood lipid indexes, such as cholesterol (CHO) content, triglycerides(TG)content, high densitylipoprotein cholesterol (HDL-C)content in serum of the rats. The experimental results showedthat Lonicera eduIis flavonoids could reduce the CHO and TG content and increased the HDL-C content, so Lonicera eduIis flavonoids had the effect of reducing blood lipid was proved. Theantifatigue effect of Lonicera edulis flavonoids was confirmed through fed Lonicera edulisflavonoids to the rats and determined the swimming time and some fatigue indexes, such asliver glycogen and muscle glycogen. The experimental results indicated that Lonicera edulisflavonoids could prolong the swimming time and meanwhile had remarkable effects onincreasing the reserves of liver glycogen and muscle glycogen. Finally, it was found thatLonicera edulis flavonoids have obvious antifatigue effect.
