Phthalocyanine (Pc) and their substituted derivatives had aroused extensive interest in the last decade, as they were widely used as photosensitizers in photodynamic therapy (PDT) of cancer or macular degeneration. Our research group is aim to developing a series of new photosensitizers for therapy choroidal neovasculadzation. The presence of both hydrophobie, hydrophilic or amphiphilic groups in the Pc ring would be beneficial for better performance in tumor selectivity. Therefore, the substitution Pc compounds with special properties and bonding properties will be synthesized and studied.In this paper, three kinds of new substitution metal phthalocyanines were synthesized and characterized. (1) Two amphiphilic tetra-α(β)-pyddyloxy phthalocyanine aluminium were synthesized. Their structures were characterized by IR,UV-Vis,1HNMR, MS, and fluorescence spectroscopy. The interaction of tetra-α(β)-AlPc(OC5NH4)4 with human serum albumin(BSA) and bovine serum albumin(BSA) were studied and compared by spectroscopic method respectively.The binding constant of two metal phthaloeyanines with serum albumins were determined by fluorescence spectroscopic method. The binding site of tetra-α(β)-AlPc(OC5NH4)4 on the HSA and BSA were determined by using Hemin chloride(HE), Ibuprofen(IB) and L-tryptophan(TRP) as ligands and competitive binding method.(2) Two hydrophilic octa-carboxylic phthaloeyanine aluminium and cobalt (AlPc(COOH)8,CoPc(COOH)8) were synthesized. Their structures were characterized by IR, UV/Vis, TG and HPLC. The interaction of AlPc(COOH)8 and CoPc(COOH)8 with BSA were studied respectively. The binding constant of two metal phthaloeyanine were determined by both UV/Vis and fluorescence spectrascopic method. The binding sites of two metal phathaloeyanines on BSA were determined by competitive binding method. According to the Foster dipole-dipole energy transfer priciple, the interaction distance between MPc(COOH)8 and BSA were measured.(3) A hydrophobic anthraquinone phthalocyanine cobalt was synthesized and characterized by elemental analysis, IR, UV/Vis, 1HNMR, HPLC. The photophysieal properties of the metal complexes were studied by fluorescence spectroscopy method.
