The crude drug Phyllanthus urinaria L, the whole herb of Phyllanthus urinaria (Euphorbiaceae, Phyllanthus), according to literature report, have a impactation on benefiting liver and clearing the toxin in human body, removing useless water and deintoxication. It also has effection on anti-tumor, healing to hepatitis, diminish inflammation, and exciting nerve centre. In this paper, the research object is Phyllanthus urinaria L. On the basis of establelishing the ultraviolet spectrophotometry method, its total polyphenols content was determined. The effective component polyphenols was extracted by microwave, ultrasonic and cellulose enzyme, respectively. The technique conditions were optimized. The content of gallic acid in the total polyphenols was measured by HPLC.In microwave and ultrasonic to be an assistant method respectively, the optimization was investigated by orthogonal designs on content of total polyphenols as a reference and different ethanol solutions to be solvent. The results indicate that the ultrasonic is better than microwave. Its technique parameters are power 200W, extracting time 90 min, proportion to solvent and material 25:1, the solvent 70%. In addition, the experiments were studied on water as a solvent by a kind of fibrin enzyme also. The optimized condition is: temperature 60℃, time 2.5hrs, pH 5.0, the content of enayme 0.5% and proportion of solvent and material 30:1, respectively.With gallic acid as the standard, the content of total polyphenols in Phyllanthus urinaria L. was established by an ultraviolet spectrophotometry. The ferrous chloride tartrate and Prussian blue method were done in mentioned way, respectively. The results shows Prussian blue method is better than that one. Its regression equation was proved to be C=10.2569A-0.07901 and the corelation R=0.9982, as well as to have a nice linearity relationship during in a range of 0~16.0μg/mL. This method is quite simple, quick and to have a sensitiveness in a certain extent.Ultraviolet spectrophotometer was used to determine the content of total polyphenols in Phyllanthusu rinaria L. Assistant extractions with ultrasonic, microwave and cellulose enzyme, was carried out respectively. The technique conditions were optimized also. These rarely were reported in relative literatures. The results supply theory basis for the industrialization in the extraction of its effective components.
