In this thesis, Duloxetine as antidepressant and Dapoxetine as male sexual dysfunction were synthesized.Duloxetine[(S)-N-methyl-3-(naphthalene-1-yloxy)-3-(thiophen-2-yl)propan-1-amine], as antidepressant, is a potent and balanced inhitibitor of serotonin and norepinephrine dual-reuptake. Duloxetine was obtained from 2-acetylthiophene through 5-step reactions including Mannich reaction, reduction with NaBH4, resolution, etherification and demethylation. The structure of the products were confirmed by NMR, MS and IR. The total yield was 35%, and the e.e value attains 93% above.Dapoxetine[(S)-(+)-N,N-dimethyl-α-[2-(1-naphthyloxy)ethyl]benzylamine] is a kind of serotonin reuptake, useding for a treatment to depressive disorder at the beginning, and it is developed to used for curing a male premature ejaculation. Dapoxetine was obtained from benzene, and 3-chloropropanoyl chloride through 5-step reactions including Friedel-Crafts reaction, reduction with KBH4, etherification, dimethylamine replace reaction and resolution. The structure of the product was confirmed by NMR, MS and IR. The total yield was 19%, and the e.e value attains 91% above.
