Male sexual dysfunction is a common male multiple diseases,including sexual libido disorder,ejaculatory dysfunctional and erectile dysfunction.Erectile dysfunction(ED)refers to men's persistent inability to maintain sufficient hardness of penis erection to obtain satisfactory sexual performance under sexual stimulation.At present,ED has become one of the important diseases that plague men worldwide.Primary gonadal dysfunction and testicular injury caused by diseases,trauma and other causes can lead to the decline of male sex hormone levels and cause sexual dysfunction such as ED.According to the “Materia Medica of Animal” records: scallops have effect of nourishing Yin and tonifying kidney,mainly used for treating Yin deficiency and strain,kidney deficiency and low back pain,spermatorrhea and other symptoms.Modern medicine shows that marine shellfish such as mussels and oysters have a good protective effect on male reproductive dysfunction.Chlamys nobilis is one of the main aquaculture economic shellfish in the southern sea area of China.Its meat is delicious,nutritious and rich in arginine and selenium those nutritional factors for improving male sexual dysfunction.In order to explore whether Chlamys nobilis has function of improving male sexual dysfunction,hemi-castrated male SD rats were used as a male rat model of sexual dysfunction in this study,and the effects of enzymatic hydrolysates and its ultrafiltration components of scallop on sexual function and serum sex hormone levels in hemi-castrated male rats were investigated,which provided a theoretical basis for exploring the physiological effects of scallops and developing scallops as dietary supplements to improve male reproductive dysfunction.The main research contents and results are as follows:(1)The edible part of Chlamys nobilis enzymatic hydrolysis(CNH)and scallop male gonad enzymatic hydrolysis(SGH)were prepared by enzymatic method,and the effect of improving male sexual function was evaluated by semi castrated male rats.The results showed that CN,CNH and SGH could significantly shorten the mount latency and erection latency of hemi-castrated rats,and increase the copulatory frequency and the attached gland organ coefficient.It could increase the serum testosterone content of hemi-castrated rats,stable the levels of luteinizing hormone and follicle stimulating hormone,and then increase NOS activity and promote the production of more neurotransmitter NO.The effect of SGH component is better than that of CNH component,followed by CN component.(2)CNH was separated by using 8 KDa and 3 KDa ultrafiltration membranes respectively,and the physicochemical properties of hydrolysates and its ultrafiltration components were analyzed.The result showed that the molecular weight peptides of <3K,3-8K and >8K were enriched after ultrafiltration.Amino acid composition of hydrolysates and its ultrafiltration components were in balance relatively,which branched chain amino acids(Val,Ile,Leu)and Arg accounted for a large proportion;furthmore,it also contains Ca,Fe,Zn,Se and other metal elements needed by human body.In addition,the TM4 cell activity experiment showed that different concentrations of ultrafiltration components had proliferation activity on sertoli cells TM4 in vitro,and the activity,increased first and then decreased,with the increase of the concentration of the sample.The effect of <3K components is better than other components.(3)Though the hemi-castrated male rats modle,the ameliorative effect of components with molecular weight of<3K,3-8K and >8K on male sexual function was evaluated.The results showed that components of <3K,3-8K could increase the catch frequency and the mount frequency significantly(P<0.05).components of 3-8K,>8K could significantly improve the catch latency hemi-castrated rats(P<0.05);<3K and 3-8K components can significantly improve the mount latency of rats(P<0.05).The <3K,3-8K,>8K components could increase the coefficient of left testis in rats significantly;and increased T content in hemi-castrated rats(P<0.01),in addition,through the negative feedback effect of testosterone regulation,LH,FSH,E2 hormones are stabilized to normal levels.The <3K,3-8K and >8K components could increase the activities of ACP,AKP and LDH in testis of hemi-castraed rats.<3K component could increase CAT activity significantly(P<0.05);<3K and >8K components could increase GSH content(P<0.05);and 3-8K component could increase SOD activity(P<0.05).<3K component could increase contents of NO and enzyme activity of NOS in penile tissue.NOS levels in the other groups were significantly lower than those in the blank control group(P<0.05);There was no significant difference in c GMP level among groups;<3K,3-8K and >8K could increase c AMP levels.Thus we seen that CNH ultrafiltration components has certain activity to improve male sexual function,and the activity of <3K is the strongest.
