| The marine fungi TS67 which has broad spectrum resistance to pathogenic fungi was screened. In this paper, culture medium and cultural conditions and the purification of the active substance was researched, and the methods of purification by adsorbing of macroporous adsorbent resin were established.The research of culture medium and cultural conditions for marine fungi TS67 active substance showed that nitrogen source presented the most significant effect on the production and that glycerol addition was beneficial. It was concluded that the fermentation activity was increased by3.1 times to 1230u/ml under the optimal conditions. The optimized medium was composed of glucose 5g/L, glycerol 5g/L, peptone 3g/L, yeast extract 1.0 g/L, and the optimized cultural conditions was that the initial pH value is 8.0, inoculation quantity is 8% ,cultural temperature is 28℃, and fermentation period is 96 hours.The research of purification of the marine fungi TS67 active substance appeared that resin X-5 is the best adsorbent; the best pH value is 5; the eluant is 50% ethanol. 50% methanol is suitable for eluting the pigment and impurity absorbed by the resin. adsorption saturation resin was flushed by distilled water, then was eluted by 50% methanol for removing the most of the pigment and impurity. Later, adsorption saturation resin was eluted by 50% ethanol at the rate of 4mL/min, then the higher active fermentation was combined and concentrated. At last, concentrated fermentation was standing for 48h and the gruff crystal was obtained.The thermal stability experiment showed that the antibiotic was instability under the 50℃or higher. The acid alkali stability showed that the antibiotic was instability in strong acidity or alkalinity. |
