Study On Supercritical CO₂ Extraction Process Of Indigo Naturalis And Pharmacokinetics Of Its Extracts

OBJECTIVE:1. By means of SFE and HPLC to establish a method for extraction and analysis of major components in Indigo Naturalis(IN). To obtain the best conditions for the extraction and analysis. 2. To establish a method for determination of Indirubin in plasma of Beagle dog and study its pharmacokinetics.METHODS: 1. To investigate the effects of factors such as amount of IN , temperature , pressure , modifier , amount of flowing CO₂ and extracting time on the recovery of Indirubin with the Orthogonal Experiment, determine the best process of extraction by means of HPLC. 2. To collect plasma samples of dog at 1, 3, 6, 10, 15, 21, 27, 33, 48, 72, 96h after ig. of supercritical extracts from IN. A HPLC method was developed for determination of Indirubin in plasma of dog. The pharmacokinetics model of Indirubin was analysed with 3P97 practical pharmacokinetics program.
