| Recent studies have given considerable evidence that phospholipids (PLs) have a positive nutritional effect on human health, such as reduction of the risk of cardiovascular disease. At the same time, PLs are natural surfactants which have many applications in cosmetic, pharmaceutical, food, and other industries. So the demand for PLs has been increasing.A method for the determination of yolk phosphatidylcholine by normal-phase high performance liquid chromatography (NP-HPLC) with evaporative light scattering detection (ELSD) was established. Yolk phosphatidylcholine was successfully separated on silica column , using isopropanal — hexance -water containing 3% acetic acid (35:65:8, v/v/v) as mobile phase. This method had good linear range and the results of several samples show that it was accurate, rapid, simple and reproducible .The method for separating and identifying the molecular species of soybean and yolk phosphatidylcholine (PC) by GC/MS was studied. The result indicates that molecular species could be separated by GC and identified by MS, and the fatty acid (FA) composition could be matched with NIST library.
