| For providing strict inspection and quarantine of kimchi for import and export, we studied the method of separating parasites eggs from kimchi and related products to prevent the Sino-Korea kimchi event from happening again. We established a series of operating instruction including the elution of 0.5% SDS solution, filtration, centrifuging sedimentation, floatation by saturated solution of NaCl and count by McMaster Counting Slides. The test of Ascaris eggs in simulative positive kimchi samples showed that the recovery rate of eggs was 35.1%. In order to identify precisely the species of parasites eggs in kimchi and other related products and distinguish from the plant-parasitizing nemotode eggs, we researched the detection technique of Ascaris egg looked as the highest polluting risk of kimchi. The specific fluorescence quantity PCR (FQ-PCR) method was established by using the Ascaris suum genome DNA as positive control,and the optimum way of breaking the eggshells in liquid nitrogen and thawing at 37℃, and the detection method of single egg. We also prepared the standard substance of recombinant plasmid including Ascaris specific gene. The detection results of the simulative kimchi samples showed that the above way and molecular identification technique can be applied to the detection of Ascaris eggs in kimchi. |
