This is the first report on the characteristic of the lentinan production of the monokaryontic mycelia of Lentinula edodes. The monokaryontic mycelium that the basidiospores from Lentinula edodes 939 bourgeon and develop could be divided into four types: type S, sticking; type A, appressed; type R, rare and thin; type H, fine hair. The dikaryotic mycelium and the four types of the monokaryontic mycelium could be cultured inthe liquid culture medium. when the monokaryontic mycelium and the dikaryotic mycelium are cultured 18d and 11d separately ,the productions of lentinan get to the fastigium: 88.85mg/100mL and 102.40mg/100mLfermentation liquid. pH-values after fermenting get pH 2.96 and 3.32.The four types of the monokaryontic mycelium could be separated into two groups based on the lentinan yield. The first group includes type S and R whose lentinan production is similar to that of the dikaryotic mycelium; the second group includes type A and H whose lentinan production is lower than that of the first group. The first group produces 8.31% more lentinan than the second group significantly.we confirm that the best induced time is 90s by UV to basidiospore of the Lentinula edodes 939 and get tow strains of highly production lentinan .The mutant (5) does not use lentinan as a C resource and mutant (10) is a biotin auxotroph. The lentinan productions of the two mutants are evidently higher than the wild type. The lentinan productions are higher 31.90% and 35.57% than that of the wild type, respectively. Mutant (5) belongs to type H fine hair and mutant(10) belongs to type S sticking, and their mycelium growth speed are slower than the dikaryotic mycelium.We research carbon resource, nitrogen resource, initial pH-value, the volume of culture medium, metal irons and get the best fermentation.
