The ultra-high pressure extraction(UPE) is a new extraction method of theessential components of plant,and the low energy consumption, the short timeconsumption and the quality of selective extraction is its major characteristic.In recentyears ,more and more scholars research the UPE in the natural medicine extractionfield. The traditional extraction methods of flavoiods from the leaves of CrataegusPinnatifida Bge.have much disadvantage such as numerous and diverse craft, the longtime and the low rate. Consequently, It is necessary for us to study extractionionmethods in search of the best extraction method of flavoniods from the leaves ofCrataegus Pinnatifida Bge.This article researched amply UPE of flavoniods fromCrataegus Pinnatifida Bge;The pharmaceutical factory discovered thatnewdosage-form of flavanoiods of the leaves of Crataegus Pinnatifida Bge. Can appearthe sedimentation after a period of depositing because macro-molecule impurity hasnot been able to clean out completely.So it is significative to research thoroughlyseparation craft parameter of total flavanods of the leaves of Crataegus PinnatifidaBge. In this paper the main contents of research and results are as follow: 1. It is the first time to extract flavoniods from the leaves of CrataegusPinnatifida Bge. by the UPE of at room temperature. Based on the study of the effectsof pressureSolvent concentration, ratio of solvent to feed material, extraction duration and extraction times on the extraction yield of flavonoids ,the optimized extraction conditions were determined by the orthogonal test design as follows:extraction solvent: 50 %ethanol;ratio of solvent(ml) to feed material(g): 45;extraction duration:10s,extraction pressure:300MPa;and 9.15% of extraction yield of flavoniods from theleaves of crataegus pinnatifida Bge was obtained.2. Comparison of UPE with other extraction method such as heat reflux,ultrasonic-assisted etc is studied, every method is operated according itself optimumconditions. The results indicate that the ultra-high pressure extraction could be cardedout not only in the shorter time but also in the higher yield.and UPE is superior to theother traditional extraction methods on the extraction of flavoniods. During theexperiment we observed that the extraction liquid of UPE is the most stable and theleast impurity in appropriate craft parameter situation ,and the extraction solution ofother methods is black,green and turbid,which showed that UPE has the quality ofselective extraction, in appropriate craft parameter situation,the impurity protein,sugar, chlorophyll to be able to reduce, which also indicated that, UPE of flavoniodsfrom Crataegus Pinnatifida Bge. is propitious to purify total flavoniods.3. It take merely 3 minutes to Complete entire UPE. the effective componentextraction ratio and the extraction efficiency enhance.4. It is the first time to uses the macroporous adsorption resin to purify totalflavanods of the leaves of Crataegus Pinnatifida Bge. the optimized separationcondition were determined by the dynamic adsorption experiment and staticadsorption experiment as follows: the total flavanone saturated adsorption quantity ofthe D101 macroporous adsorption resin: 78mg/g;the water elute quantity : 8BV;theethanol elute quantity is 4BV;adsorption speed of flow is 3BV/h, desorption speed offlow is 2BV/h;the eluting sequence is that with the fuicl of 10% ethyl alcohol eluting,abandoned, the fluid of 70% ethyl alcohol elutes is collected, concentration anddryness, and total flavanods content may achieve above 80%.5. The effect of the different concentration ethanol as elution solvent onflavanods was studied. And the experimental results showed that the differentconcentration ethanol as elution solvent, the kind and content of the flavonoids areobviously different. If taking 2"-O-Rhamnosylvitexin as the goal, the eluting sequenceis that with the fluid of 30% alcohol eluteing abandoned, the fluid of 50% alcoholeluting is collected, concentration and dryness:6. The antioxidant ability of 7 kind of Chinese traditional medicine were studied.The research results indicated that the leaves of Crataegus Pinnatifida Bge take on thehigh antioxidant ability.At present the leaves of Crataegus Pinnatifida Bge are rich inchina, but few leaves of Crataegus Pinnatifida Bge are empoldered and utilized,therefore it's significative to thoroughly research the leaves of Crataegus PinnatifidaBge.and its application.7. The flavonoids content in the leaves of Crataegus Pinnatifida Bge. Of differentgrowth stages are studied.The contents of flavonoids were determined by HPLC andUV-spectrophotometry. At different growth stages, the content of total flavonoids and2"-O-Rhamnosylvitexin were also the highest at August;afterwards, the flavonoidscontent decreased, so August was appropriate time in order to obtain flavonoidscomponents from the leaves of Crataegus Pinnatifida Bge. Its antioxidant ability alsopresents such tendency.
