Chemiluminescence has been exploited in a number of analytical applications ,owing to its high sensitivity, wide linear range, low detection limits, rapidity, simplicity and relatively low cost. It is well known that pharmaceutical analysis has taken great important role in clinical, pharmacokinetic research. It is not only of great theoretical meaning but also of application value that chemiluminescence is applied in the pharmaceutical analysis. This thesis was composed of two parts. In the first part, a review was presented. Firstly, all kinds of CL reaction and their applications in pharmaceutical analysis were briefly described. .Secondly, An overview on the combination of CL with other analysis technology was presented .From the overview , we know that CL is a powerful trace-amount analytical tool in the future, especially in pharmaceutical analysis. The second part is research. According to CL theory , the CL behavior of gallic acid, cimitidine and famotidine was investigated. Rapid, simple and sensitive, flow-injection CL methods for the determination of the above pharmaceutical preparations were established. And CL mechanism was also discussed.1. Based on the reduction of hexavalent chromium by gallic acid to trivalent chromium which in turn catalyzed the chemiluminescence of luminol with hydrogen peroxide, a rapid, sensitive and new method for the determination of gallic acid was established. Linearities for gallic acid was found in the range of 2×10-95×10-6g/mL with the detection limit of 1.2×10-9g/mL. The relative standard deviation(n=11)for 1.0×10-7g/mL gallic acid was 2.1%. This method was successfully applied to the determination of gallic acid in Jianming Yanhou tablets.2.A novel chemiluminescence method for rapid determination of cimetidine combined with flow injection analysis was presented in this paper based on the CL reaction of cimetidine and hexacyanoferrate(Ⅲ) in sodium hydoxide-sodium hydrogen carbonate medium. A short reaction coilwas designed in order to catch the greatest CL signal. The proposed method had a linear of 5 X 10"7 1X HrVmL wjth a RSDof 1.8%( 5 X lG*g/mL, n=ll). The detection limit of this method was l.lX10'7g/mL. The method was successfully applied to the determination of cimetidine in tablets.3. A new sensitive method for determination of famotidine with flow-injection CL was presented. It was based on the CL emitted during the oxidation of famotidine by N-bromosuccinimide in alkaline condtion in the presence of Rhodamine B as energy transfer. This method was greatly enhanced by cetyltrimthylammonium bromide. Various factors influenced the CL system were investigated carefully. The possible mechanism of the proposed method was also studied preliminarily. The detection limit was 5 X 10"8g/mL, the linear range of determination was 1X10'7 g/mLl X 10'5gML and the RSD of 2.1 %(5 X 10"7 g/mL, n=ll). The recommended method was successfully applied to the determination of famotidine in tablets. | | Keywords/Search Tags: | chemiluminescence, flow injection, gallic acid, cimetidine, famotidine, pharmaceutical, hexacyanoferrate(Ⅲ), N- bromosuccinimide, Rhodamine B, cetyltrimthylammonium bromide | PDF Full Text Request | Related items |
