| In this paper, the reflux of ethanol and the technique of separation with silica gel were used to extract and separate ursolic acid from Forsythia suspensa (Thumb.) Vahl. The optimal condition of reflux extraction and the parameters of adsorption were thoroughly studied.By orthogonal design, the optimal condition of is: the temperature is 85 ℃, the ethanol concentration is 75%, liquid to solid radio is 10, extraction time lh, extracted 3 times, the yields of ursolic acid were 6.182mg/g; By uniform design, the optimal condition of is: the temperature is 85 ℃, the ethanol concentration is 75%, liquid to solid radio is 7, extraction time lh, extracted 2 times, the yields of ursolic acid were 6.154mg/g. The method of extraction of ursolic acid has been studied by cellulase. The results indicate that pH of enzyme solution is 4.0, temperature of cellulase degradation is 40 ℃, time of cellulase degradation is 1.5h, the amount of enzyme is 7.5mg/g Forsythia suspensa (Thunb.)Vahl and the yields of ursolic acid were 6.313mg/g. Comparing two extraction technologies under the respective optimization extraction conditions, we found that the yields of ursolic acid by cellulase were higher a bit than the... |
