Forsythia suspensa stems is the dried stem of the Forsythia suspensa in the family Oleaceae.Forsythia suspensa is an authentic medicinal material in Shanxi,and its fruits are often used as medicine.It has the functions of clearing heat and detoxifying,reducing swelling and dissolving,and has been used in the history of Chinese medicine for more than 2,000 years.Forsythia suspensa is widely distributed in eastern Asia and many parts of Europe.Domestically,it is mainly distributed in western China,northern China,and central China.Forsythia suspensa stems is the stalk of Forsythia suspensa,and its related literature is less documented.Among the contemporary works The Chinese Materia Medica and Chinese Medicine Dictionary contain that Forsythia suspensa stalks and leaves have the characteristics of bitterness and coldness,and its main function is to clear heat and detoxify,and treat the heart and lung heat.In the process of Forsythia suspensa planting and wild breeding,in order to improve the seed setting rate of Forsythia suspensa fruit,Forsythia suspensa plants are pruned every year in the main Forsythia suspensa production area,and a large number of Forsythia suspensa stems and leaves are discarded,causing huge waste of resources.Earlier study by this research group shows that Forsythia suspensa stems mainly contain phillyrin,(+)-pinoresinol-β-Dglucoside,(+)-epipinoresinol-4ˊ-O-β-D-glucoside and so on.At the same time,its extract has a strong antibacterial effect on Staphylococcus aureus,and the antiseptic activity is better than the Old Forsythia.At present,there are many research reports on the use of Forsythia suspensa leaves.Wang Qingbo of our research group transformed phillyrin into phillygenin through enzyme fermentation of Forsythia suspensa leaves,and established the preparation method of phillygenin through separation and purification technology.In this paper,the Forsythia suspensa stalk is fermented,and the best fermentation conditions are screened.In addition,the composition differences before and after fermentation were compared and analyzed,which provided a basis for the development and utilization of Forsythia suspensa stalk.The specific research content is as follows.1.A HPLC method for the determination of phillyrin,arctigenin and phillygenin in Forsythia suspensa stems was established and verified.The contents and changes of three lignans,phillyrin,arctigenin and phillyrin,were compared and analyzed with the stems of Forsythia suspensa collected in different periods from April to October.At the same time,the contents of lignans in different parts of Forsythia suspensa plants were compared and analyzed.The experimental results show that the content determination method established in this study has good stability,repeatability and accuracy,and can be used to determine the content of phillyrin,arctigenin and phillygenin in Forsythia suspensa stems.Forsythia suspensa stalks contains rich phillyrin components,and the content of phillyrin in Forsythia suspensa stalk collected in April and August is higher,1.68% and 0.72%,respectively.In addition,the determination results of lignans content in different parts of Forsythia suspensa plants show that the content of lignans in the leaves of Forsythia suspensa was higher,and the contents of Forsythia fruit and the stem of Forsythia suspensa are similar.2.By using the method of solid-state natural fermentation of Forsythia suspensa stem and taking the content of phillygenin as the index,the influence of fermentation temperature,water content of raw materials and fermentation time on the production of phillygenin was studied through single factor condition experiments and orthogonal experiment.Finally,the optimal conditions for natural fermentation of Forsythia suspensa stalk were determined,and repeated verification experiments were performed.The results showed that the best natural fermentation conditions of Forsythia suspensa stems were as follows: 75% moisture content of raw materials,35 ℃ fermentation temperature,and 3 h fermentation time.Three replicates displayed that the average content of phillygenin in the fermentation product was 3.83 mg/g and the RSD of the parallel experiment was 1.10%,which indicates that the fermentation process is stable and reliable.In addition,the study found that the composition of Forsythia suspensa stems changed greatly before and after fermentation,and further research is needed.3.UPLC and LC-MS analysis methods are used to analyze and identify the chemical components before and after fermentation of Forsythia suspensa stems.Column chromatography was used to isolate and purify the specific chemical components produced by the fermentation of Forsythia suspensa stems.The results of the study found that not only the content of chemical components was changed,but also produced new chemical compositions,which were initially verified as pinoresinol monomethyl ether and matairesinol.
