| The paper studied the optimum extraction condition of the fat-soluble pigment from Tagetes patula L. petal by means of single factor experiment and orthogonal experimental design. The result showed that the optimum extraction condition was 45℃,1:30(g/ml),4.5h with ethyl acetate. And discussed the stability of the fat-soluble pigment from Tagetes patula L. under the different conditions such as light , temperatures , pH, oxidants , reducing agents , metal ions , potassium sorbate , saccharides by studying the vary of feature absorbance. Results showed that it was stable when heating, saccharides, oxidants, preservatives or reducing agents of low consistence were applied to its solutions respectively. It was sensitive to light and unstable when pH>7.0 and presence of Na+,Mg2+ could lead to its loss. Then studied the separation and pureness method of crystallization on the fat-soluble pigment from Tagetes patula L. and compared results of some different methods of crystallization. Finally, the method of three-solvent crystallization is found, which could obtain red needle of the fat-soluble pigment from Tagetes patula L. .Meanwhile, a theory basis for this method was advanced. The result showed that the optimum extraction condition was 50℃, 4h with 60% ethanol concentration. The pigment was unstable when sunlight or temperature>60℃. Antioxidative activity of the fat-soluble pigment from Tagetes patula L. was studied, and the antioxidaive activity of the pigment with three kinds of amino acid were discussed. The results showed that the fat-soluble pigment from Tagetes patula L. had some antioxidative effect. Tyrosine had synergistic antioxidative activities with the pigment. And it had similarly antioxidant as VE. |
