This work is rest on the studying and investigating the separation and purification processing of a new antibiotic that is produced in zymotechnics, and the key factor that can destabilize the AGPM, and a suitable separation and purification process that should be established.Because the giant biotic molecular, AGPM, has some physiological and pharmacological activity. In order to keep its activity, we should treat it under the processing in according to its nature. This work has discussed the stability of AGPM to hot, acid and alkali, enzyme, reagent et cetera in processing, induced the active law of AGPM to those, ascertain the separating and purifying process and technological condition.With the technology of TLC, HPLC, in vitro antibacterial efficacy assays to track the variety on the components of AGPM we have seen the course of disintegration of AGPM more clearly than ever, provided some information for more research work.This work has proven the law in the stability of AGPM. AGPM in the broth is not a stable component to higher temperature. Its extreme operating temperature should be at 50 ℃. At 75 ℃ AGPM will quickly lose its antibacterial activity. The pH of its broth is the most crucial factor on its stability. In the separating and purifying process the first operation should modify the broth pH to 9. Otherwise, when it is at acidity or when there are some reagent-obtaining electrons AGPM will vary quickly lose its antibacterial activity. And when it is at acidity alkalescence or when there are some reagent-offering electrons AGPM will be very stable. There are some alternative effects on temperature, pH, removing of protein. When it is under the alkalescency condition its stability is batter than acidic, when protein is removed from its broth its batter than not. The optimized condition for separations and purification are 30℃, pH9, non-protein broth.
