Modern pharmacological studies have proved that Kudingcha has many medicinal activities. Whose extract is being used for the preventing and treatment of heart disease, multiple sclerosis, cancer, tumors, autoimmune disease, inflammation, high blood pressure and high blood sugar. These activities mainly result from many aromatic, terpenes and flavonoids compounds in kudingcha. So the studies on extraction and chemical constituents of kudingcha is of great theoretic and practical significance.The comparative studies of four kinds of extraction aromatic methods are reported. The results showed that ultrasonic wave assistant distillation extraction is the best way. By this method and GC/MS analysis nineteen compounds were extracted and isolated, including alcoholates, esters, sesquitones and terpenes.Six approaches were used to extract flavonoids from Kudingcha. According to the typical colorimetric reaction that maximum absorbency can be obtained at 510nm when the chelate formed by the combination of flavonoids and metal ion. spectrophotometry was used to determine the total flavonoids content calculated by Rutin standard. The analytical results showed that microwave and ultrasonic wave extraction are best way. But microwave extraction is difficult for industrialization, so design orthogonal experiments to discuss the best extraction. By single factor and orthogonal experiments the results showed that the optimal conditions for ultrasonic wave extraction were 40%ethanol, ratio of material to solution (w:v)1:30, andextracted for 30 min in ultrasonic wave; and for ethanol extraction were 80%ethanol, ratio of material to solution(w:v)l :20, and refluxed for 3h.Kudingcha was extracted according to above ultrasonic extraction conditions, isolated by polyamide and silica gel chromatography. Silica gel chromatography column was eluted with a CHCl3-MeOH gradient, in CHC13-CH3OH (100:25) gradient a light yellow powder was obtained It maybe 2-Deoxy-D-galactose identified by IR. Polyamide chromatography was eluted with different percent ethanol (10%, 30%, 50%, 75%, 95%). Eluted again and again, in 30%ethanil only a yellow powder was obtained.
