The natural fluorescence of biological molecules in marine can reflect the productivity of phytoplankton in the upper-ocean and ecosystem structure.The method of doing research on marine ecological processes through the changes of fluorescence properties of Marine biological molecules has advantages of high selectivity, high information and so on.Therefore, the study of three-dimensional fluorescent properties of marine biological molecules is very necessary both in the study of the structure of fluorescent substances for laboratory and in the research of ocean situ online fluorescence detection.The dissertation presents a method to study the three-dimensional fluorescence spectra properties of biological molecules through extracting characteristic value three-dimensional fluorescence after comparing three-dimensional fluorescence spectra of various analytical methods research status. 99 groups of exprement program are designed to do the research on fluorescent properties of marine biological molecules which are carried out at various environmt conditions such as various pH values, the influence of various metal ions and the influence of pressure changes.The results show that the three-dimensional fluorescence characteristic of tryptophan is related with solution of pH environmental. Certain metal ions can make tryptophan fluorescence happen to quench, and the quenching is related with solution of pH environmental and the ion concentration. Tryptophan fluorescence intensity is increasing with pressure increase. And its three-dimensional fluorescence spectra exists red shift. The study of tryptophan fluorescence fill the gaps, make up for the limitations of the traditional spectral analysis method and provide some reference value for the research of the ocean dissolved protein and amino acids and the study of tryptophan as exogenous or endogenous probes to explore protein conformational changes and so on. The dissertation includes six chapters:Chapter 1 elaborates the background and signification of the study. The current developments in correlative field are introduced in this dissertation, too. This chapter details the strengths and weaknesses of current methods, develops a new exprement program to analysis the three-dimensional fluorescence characteristic of tryptophan.Chapter 2 describes the principle of three-dimensional fluorescent spectrometry analytical method. On this basis, the chapter discusses different analytical method in detail.Chapter 3 does research on three-dimensional fluorescent characteristics of tryptophan on the conditions of different pH value.Chapter 4 does research on three-dimensional fluorescent characteristics of tryptophan on the influence of various metal ions.Chapter 5 does research on three-dimensional fluorescent characteristics of tryptophan on the influence of pressure.Chapter 6 is a summary and a prospect of the entire dissertation.
