Plain prinia(Prinia inornata)and yellow-bellied(P. flaviventris)prinia,subjected to Passeriformes Cisticolidae, are the relative species in modality and zoology. Both of them are monomorphic birds and it's impossible to process the sex identifiation well by basing on exterior characteristic and behavior. Because sex is one of the most important characteristics of birds, and male and female birds generally appear the diversity in physiology, behavior, zoology etc.,it's specially important to find the right sex identification method for the zoology research on the two species and other monomorphic birds.This research is to feed a number of plain prinia and yellow-bellied prinia in cages, and identify their sex by CHD gene.By separately recording their vocalizations in the condition of knowing their sex, find vocalizations and behavior diversity between male and female. At the same time, try to identify the sex of the two species according to their vocalizations and solve the problem of identifying the sex of monomorphic birds in field. The main research and results are as follow:1.Molecular sexing of plain Prinia and yellow-bellied prinia:We applied method of CHD gene to sex plain prinia and yellow-bellied prinia. We find that:1)Primer 2550F/2718R was not applicable for sexing this two species but primer P2/P8 did;2)Results of molecular sexing matched entirely with anatomizing results;3)DNA can be also extracted from primaries and secondaries of live individuals.2.Vocalization differences in sex of plain prinia and yellow-bellied prinia:1)Vocalization differences in sex of plain prinia:The configuration of plain prinia' vocalization is briefness,such as type of single syllable repetition and type of serial phrase stable.The female ones own 3 kinds of call,all of them are type of single syllable repetition;the male ones own 12 kinds of vocalization, the types of syllable variety is diverse.The typical song types 2 and 12 can be used to identify the sex of plain prinia in field.The highest frequency and main peak frequency and syllable duration of the female are hyper than that of the male.So the female must pay out more energy on their call.For this reason, their timbre may more euphonic by ear.2)Vocalization differences in sex of yellow-bellied prinia:The configuration of female yellow-bellied prinia is terseness,one call belongs to the type of single tweeting call,the other two ones belong to the type of single syllable repetition.All of them are gently by ear,and the singing rate is well-proportioned.There's no apparently seasonal variety among the female's call.The configuration of the male ones is diverse, containing type of single syllable repetition,type of single syllable song, type of two syllables repetition and type of serial phrase stable.The male's vocalization is very fair-sounding,and has apparently seasonal variety, possess the typical song in breeding season.The bandwidth syllable duration of the female are subter than that of the male. The typical song typesâ…¡andâ…£can be used to identify the sex of yellow-bellied prinia in field.We concluded that primer P2/P8 was applicable to realize rapid sexing for yellow-bellied prinia and plain prinia and might be applicable for the sexing of the birds of Cisticolidae.The vocalization of two species have distinct differences in sex,which main represent in the configuration and mode of vocalization.We speculate that the differences of male and female's vocalization might have their own ecological meanings.
