Low temperature is considered one of the most key environmental factors that restricts growth and distribution of plants and finally results in poor crop productivity and hurt in photosynthetierate.Caragana korshinskii Kam distributes widely over the fixed and semi-fixed dune of the steppe desert and the typical desert zone. It has an excellent water absorption and conservation capacity, and adapts to low temperature. The current studies are mainly focused on its physiological, ecological and biological characteristics, little is known about its molecular mechanism.The cDNA from the seedlings of Caragana korshinskii Kam treated under 4℃was used as tester, and the cDNA f rom the seedlings in normal growth condition used as driver, suppression subtractive hybridization (SSH) was established for isolation of differentially expressed transcripts responding to low temperature stress. The completed library contained 1,318 clones, and all clones had about 250 - 750 bp inserts. The clones were sequenced and analyzed by BLAST. Finally, 238 Contigs and 584 Singlets were obtained. Totally 822 UniESTs were recovered in this library.Caragana korshinskii Kam is rich in polysaccharide. A modified method was used to extract high quality of RNA in this study.
