Low temperature is the important factor affecting plant growth and development,which is also one of the main factors leading to a decrease in the yield of crops in agriculture,and affecting the agriculture industry in China as well as the whole world.Therefore,to improve the tolerance of crops in low temperature using transgenic techniques has great significance.This thesis aims to study the function of arabidopsis actin depolymerizing factor ADF5 in plants within the low tempreture.We use transgenic overexpression plants to study the location of ADF5 in plants'cell and its relationship with microfilament, meanwhile we realize the function of ADF5 in low tempreture stress. We also study the expression mode of this gene in low tempreture stress and analyze its anti-tolerance mechanism in order to unveil the the function of arabidopsis actin depolymerizing factor ADF5 in plants within the low tempreture.We made use of the cloned gene ADF5,ADF5R,T10A,T10D and plant expression vector 1300 to structure plant expression vector 1300-ADF5-RFP, 1300-ADF5R-RFP,1300-ADF5-T10A-RFP,1300-ADF5-T10D-RFP.We leaded the plant expression vector 1300-ADF5-RFP,1300-ADF5R-RFP,1300-ADF5-T10A-RFP, 1300-ADF5-T10D-RFP into Arabidopsis within agrobacterium-mediated genetic transformation system, the above vectors filtered by hygromycin,1300-ADF5-RFP, 1300-ADF5R-RFP,1300-ADF5-T10A-RFP,1300-ADF5-T10D-RFP got T1 generation,1300-ADF5-T10A-RFP got T3 generation, meanwhile, fluoroscopic examination proved that the plant expression vectors had been integrated into arabidopsis FABD2 genome.Experimantal results show that 1300-ADF5-T10A-RFP has impact on the growth of trangene arabidopsis. Speculating 1300-ADF5-T10A-RFP affected the shape establishment of plants initially, like the following results.1300-ADF5-T10A-RFP T3 generation of arabidopsis shape observation:transgene T3 generation plant has sparse roothair and warp; compared with dense and straight roothair,1300-ADF5-T1OA-RFP T3 generation of arabidopsis divided into 3 batches and 12 strains for each batch compared with 12 strains arabidopsis 1300 empty vector during the contrast initial flowering time, the observation show that the flowering time of transgene arabidopsis postponed 7-8 days than the comparison. Compared with the wild type, the pollen tube of 1300-ADF5-T1OA-RFP T3 has two pollen tubes, one long and one short.
