At present, many researchers at home and abroad have studied the turtles' phylogenetic relations, using 18S rRNA,28S rRNA,12S rRNA and so on.However, there are no reports about the research on turtles'phylogenetic relations by Cytb gene.So this paper made the Cytb gene as a control area to study the molecular classification of six kinds of turtles, in order to provide more reliable and accurate informations for phylogenetic.Abstracted DNA from muscles of six turtles, and amplified DNA by PCR. Then, put these DNA sequences into analysis by the gene analysis software MEGA 4.0 and Clustalx 4.8, and constructed the phylogenetic tree. The analysis results are as follows:(1) in the 1249bp base pairs of six turtles, the content of identical sites is 813bp, including TT 208bp, CC 244bp, AA 251bp, GG110bp; The content of transitionsal pairs is 170bp, including TC 101bp, AG 69bp; The conent of transversional pairs is 163bp, including TA 42bp, CA 49bp, CG 38bp; The quotient of the transitional and transversional pairs is 1.0.(2) The content of the conserved sites and variable sites between the Chinemys nigricans and Chinemys reevesii is 1142 and 7, respectively. They have the most conserved sites, compared with other four turtles. This shows that they have a close relation of affinity; The content of the conserved sites and variable sites between the Annamemys annanmensis and Mauremys mutica is 1036 and 106, respectively. They have the most conserved sites, compared with other four turtles. This shows that they have a close relation of affinity; The content of the conserved sites and variable sites between the Ocadia glyphistoma and Trachemys scripta elegans is 1142 and 7, respectively. They have the most conserved sites, compared with other four turtles. This shows that they have a close relation of affinity.(3) From the constructed phologenetic tree, it shows that Trachemys scripta elegans is at the bottom of the tree and has a close relationship of affinity; The Chinemys nigricans and Chinemys reevesii are in the middle of the tree and have a close relationship of affinity; The Annamemys annanmensis and Mauremys mutica are at the top of the tree and have a close relationship of affinity.
