Electric-Field of Needle-Plate Corona, which is applied to a wide range of air purification, virus inactivation and polymer surface modification, has become a great success. In this thesis, a pin-plate corona field is successfully constructed to be testing equipment for genetical modification. It not only broadens the research field on Electric-Field of Needle-Plate Corona, but also enriches the methods for transgenic technology.In the research process dealing with the mutation effect which is caused by treating the hedysarum scoparium with Electric-Field of Needle-Plate Corona, the thesis author discovers that the treatment conditions of Electric-Field of Needle-Plate Corona have certain influence on the germination rate of hedysarum scoparium. As is proved in the experimental results, the germination rate can reach relatively high values if the processing time is 90min and the voltage from 14kv to 15kv.It is shown in the process studying the inducement of hedysarum scoparium callus that the induction media rate is fairly high and the callus is of the best quality when the concentration of induction culture medium which contains 6-BA is around 1.5mg/L and that of the 2,4-D is about 2.5mg/L. The thesis author also discovers that different treatment conditions have certain influence on the survival rate of callus when treating it with Electric-Field of Needle-Plate Corona. The callus should not be dealt with in a long time when the voltage is 15kv.In the course of transforming plasmid pBI121/Gy3 into hedysarum scoparium callus mediated and recomposed by Electric-Field of Needle-Plate Corona, the experimental results show that when treating the callus under the voltage of 14kv for about 90min, the Gus instantaneous expression rate of the callus can reach 52% and the survival rate of the callus is also relatively high, at about 80%. Therefore, the best treatment condition for DNA callus transformation mediated by Electric-Field of Needle-Plate Corona is under the voltage of 14kv for about 90min.To transform the hedysarum scoparium callus dealt with Electric-Field of Needle-Plate Corona into the regeneration medium with different hormone proportion, the thesis author gets the conclusion from the experimental result of the callus budding differentiation rate that the time for the hedysarum scoparium callus differentiation is relatively short and the way the buds differentiated grow is the best when MS culture medium which contains 6-BA is 2.0mg/L and that of the NAA is 0.2mg/L.
