Leaf is the basic nutritional organs of higher plants. Regulatory mechanism of leaf development is very complicated. The development of leaf is under the control of both environmental factors and internal genetic factors.The identity of leaf polarity determines leaf flatness development directly,and abnormal polarity usually results in the abnormal leaf flatness,subsequently affect on many physiological functions,such as photosynthesis,transpiration and anti-stress.Hence,understanding the genetic mechanism of leaf polarity or leaf curvature has the theoretic significance for controlling leaf form and shape,and for the leaf development theory.In this paper,we studied abnormal leaf development mutant gene function by a method of being a genetic.The mutant gene was cloned by use of map-based cloning method from Arabidopsis. The main findings and conclusions are as follows.1) Morphological observation of mutant:leaf of mutant in both horizontal and vertical coils to varying degrees,it was more great degree of curling at an early stage.By the late development of curly leaf significantly was smaller.At the same time,leaf angle became larger.It has a node during flower development there,and bending.Silique also changed the perspective.2) Studied the response of the hormone in mutant. Respectively,mutant and wild type grown at the different concentration of hormone.We found that Mutants were more sensitive in high concentrations of IAA treatment on the inhibition of IAA than the wild type3) Analysis of mutants on the genetic:The mutants was hybridized with the wild-type,self-hybridized.F1 populations were the wild-type traits.Characters appeared isolated in F2.The plant separation ratio of wild-type and mutant traits was 3:1 in F2 populations.It showed that the mutation was a single recessive gene mutation.4) By use of map-based cloning technology,mutant gene has been initially located in the third Arabidopsis chromosomes, with molecular markers closely linked GAPAB.
