Surface plasmon resonance (SPR) is one of the most effective methods for analysis of interaction between biomolecules. There are some unique characteristics of freelabel, real-time and quick response on surface for SPR sensor, which plays an important part in analysis of biomolecules'interactions and supplies ample information about some interested materials in the complex samples on some concentration level and/or specialization, affinity, kinetic relation. SPR was advanced as concept until in the 1960s. However, it was get more attention of researchers as a real spectral technology only recent several years. Today, the applications of SPR technology get huge advances that have already covered hot fields including pharmacy, chemistry, engineering, environment and biology. It could combline with mass spectra, micro-fluidics and nanoparticles to develop each advantage in order to get more amazing effects. It also made positive action in the development of chemistry, physics and engineering that the methods of bioanalysis and instrument/technology for SPR had progressed.There are four chapters in thesis. The first chapter introduces the types of SPR sensors; according to SPR'configurations, it was classified with prism-coupled sensor, grating-coupled sensor and fiber optic-coupled sensor; based on SPR's detection mode, it also was classified with wavelength-modulated sensor, angle-modulated sensor, poloarization state-modulated sensor, intensity-modulated sensor, phase-modulated sensor. In the second chapter, we introduced a wavelength-detected SPR instrument that it was homemade by ourselves. LED is used as light-source, which goes through two lenes and a polarized len to become polarized-parallel beam. The incident angle of light-source was fit and Kretschmann-brism was used, that made the beam inflected the thin gold-film surface, so there is happened attenuated total reflection. Light-wave is coupled with metal surface plama to occur resonance action. Exit-beam optical system that is detection system made the reflectical signal focused on optical fiber by convex len to enhance signal and increase sensitivity. Charge coupled device (CCD) is used as detector to mearsure the optical signal. And there is any the mobile component in the instrument.The third chapter, we employed wavelength-detected SPR biosensor to measure the interactional statement between different GPX mimics and its substrate GSH. SPR biosensor is unlabeling to detect the action. It is easy and time-saved that GSH is directly immobilized as sense surface on sensor and also GPX mimics'substrate. We have got the results that kinetic curves, adsorption rate constants, disossiation rate contants and accossiation constants about interaction between 2-TeCD/2-SeCD/6-CySeCD and GSH. Comparing with the three GPX mimics'accossiation constants, we got the conclusion that binding ability of 6-CySeCD and GSH is the highest, and 2-SeCD's is relative weak.In this section, traditional method of direct measurement was used to abtain preliminary information of interaction between GPX mimics and GSH on SPR sensor surface, which is the based research of sandwiched and signal-amplified methods. However, the above-mentioned study supplies relational information that GPX mimics become artificial GPX drugs.The fourth chapter, we used the same device to measure the hybrication of DNA of Caspase-3 on SPR biosensor. The specificity of avidin- biotin was employed. One strand of Caspase's DNA fragment was added biotin as bio-probe, and the other strand DNA is freeable in the solution as analyte. The limit of detection is 14 nmol/L without signal amplified. Based on this detectional limit, we've got the limit the detection with silica-coated CdTe QDs amplified, which is 26.2 pmol/L. The research of methods for detecting Caspase on the SPR sensor surface is very convenient and easy and could get good effect.
