The technology of surface plasmon resonance was applied in the equation of two different substances, some conclusions were educed, at the same time, the detecting techniques and the potential application of the micro total analysis systems were also discussed in this thesis. The content of this thesis were divided into five chapters and listed as follows:1. The development of the history of surface plasmon resonance and it's physical-optics principles, characteristics were discussed in the chapter, and the current development instance of SPR instruments were referred simply also. At last, the applications of SPR technology were concluded in 11 fields, and the newest development and trends of SPR technology were also introduced.2. Under introducing the general aspects and history of the micro total analysis system, this paper summarized the detecting techniques and discussed the trends and its potential application.3. The interactions of ss-DNA and SSB were studied in the third chapter. First, the interactions of ss-DNA connecting to Au chip by static affinity and SSB were studied, then the roles to the reaction of the Mg2+ and Na+ in the solution were also studied. The second, an ss-DNA gold chip was prepared based on self-assembly of the thiol-derivatized oligonucleotide, and used for the interactions of single-stranded binding protein (SSB) by surface plasmon resonance (SPR). The results showed that SSB binded ss-DNA with high specificity, and the relative signal of SPR response is proportional to the concentration of SSB in the range of 0.1~100 ng/mL with a detection limit of 0.07 ng/mL under the condition of the flow rate is 17.12μL/min and the pH of the buffer Tris-HCl is 7.4.4. The interactions of phenanthroline with iron(II) ion were studied by SPR technique. Both the SPR response rules and the adsorption to the gold surface of phenanthroline and iron(II) are discussed respectively. The results showed that SPR angel changed very little when the concentration of phenanthroline or iron(II) changed from 10-6 mol/L to 10-4 mol/L, but the combination of phenanthroline with iron(II) produced great SPR response. Just for this reason, some meaningful information about the interaction process can be gained by SPR technique, such as the combination ratio is 3, the combination constant is 1.79×1017 L/mol.5. In the fifth chapter, BSA was immobilized on the SPR-2002 gold chip by a dual amine bond successfully, and the optimized experiment conditions were proved.
