The β-Ocimene Synthase Gene Cloning And Its Over-express Vector Construction Of Plant

Plant communication means the signal inter-space transduction and the signal acceptor's physiological responses between the individual plants and plants with the herbivores.Plant after the attack by herbivores will produce the volatile organic compounds,such volatile compounds play an role on signal inter-space transduction.Most of them are low-molecular-weight Terpenoids,including monoterpene,sesquiterpenes and diterpene. E-β-Ocimene is one of the monoterpene volatile organic compounds which found in recent years is a significant plant communications signal molecule.Now that we found after herbivores-attacked,many plant species such as:corn,Lima beans,potato,tobacco, arabidopsis,the mediterranean cotton and tomatoes will release theβ-ocimene.To further clarify theβ-ocimene's function as a signal molecule.we According to the cDNA sequence of AtTPS03 gene in GenBank,a pair of primer was designed and synthesized.Using the total RNA from JA treated Arabdopsis thaliana seedling as the template,the full length cDNA of AtTPS03 gene was amplified through RT-PCR and was clone it into pMD18-T vector,DNA sequencing indicated that the cloned fragment showed 100%identity to the sequence of AtTPS03 gene in GenBank.,the the cloning fragment was ligated into pCAMBIA1303 and we got the AtTPS03 gene express vector of pCAtP35S regulated by 35S promoter.And through the GUS gene instantaneous expression proved this over-express vector have been constructed succssefully.The purpose of this study is to promote theβ-ocimene Synthase gene over expression by Arabidopsis.So as to further clarify theβ-ocimene's functions as a signal molecule and lay the foundation for understanding the signal inter-space transduction and the signal acceptor's physiological responses between the individual plants and plants with the herbivores machanism.
