As prokaryote, cyanobacteria show special character in photosynthesis, nitrogen fixation, metabolization and evolution. Because of simple genome, it's good for the operation of gene engineering. Particularly in recent years, cyanobacteria have gradually become a very good acceptor system by fast development of gene engineering.Photosynthesis is the most important chemic reaction in the world, and it is the ultimate energy source of any living organisms. Photosynthetic regulation is also a hot pot in cyanobacteria gene engineering, apcF is a important subunit in light reaction center. apcF is encodingβ18.5 amino acid ,β18.5 which is a part of allophycocyanin(APC).β18.5 can transfer light energy in APC. As we know, the energy transformation process is PE→PC→APC→Chla in photosynthesis. From the process APC→Chla, the energy transformation is to PSⅠa nd PSⅡindependently. In particularly, apcF is supposed a key function gene of energy transformation from APC to PSⅡ.In this experiment, the reconstructed plasmid vector is transformed in Synecocystis sp.PCC6803 by nature transformation. The reconstructed plasmid contained apcF homologous fragment,reporter gene(Kanamycin resistance gene).The Kanamycin resistant transformants are obtained by Kanamycin screening.The apcF inactivated mutant are confirmed by PCR.In the experiments, from the assay of absorption and fluorescent spectrum under room temperature, and a series of physiological and biochemical index, we have not only analyzed the influence to phycobilisome and cyanobacteria pigment, but also put forward to the possible mechanism of catching light energy when parts of phycobilisome is destroyed and give researching into the mechanism of photosynthesis.
