Where the human being came from,how they have been origined,how the races, nationalities and different groups were differentiated and developed,these are the questions that anthropologists have long been exploring.For a long time, anthropologists analyzed origin,differentiation of human and genetic relationships between different races,ethnic groups and populations from the aspects of archeology, linguistics,ethnology and humanities.However,the studies of molecular genetics indicates that all the differences between races,ethnic groups,even among indivisuals are the differences of molecular level in essence.Analyzing the essence of differences in genetic relationships between races,ethnic groups on the DNA molecular level, finding out the reasons due to the differences,understanding genetic structure,origin, migration of every ethnic group and population,are the keys of exploring the human evolution and development fundamentally.Moden Lopnur people living in Lopnur county of Xinjiang who consider themselves as Uyghurs are still remaining some distinctions from other Uyghurs on living habits,language,apperiance and clothing. The unique life style of Lopnur people has aroused interest of many researcher's from in and out of the country,and their history,traditions,dilects are hot research topics. But,there has not been any study on genetic diversity of Lopnur people.Therefore, we studied the mtDNA and Y-chromosome diversity of Lopnur people from the genetics perspective intially.Objective:Carrying out fundamental studies on the origin of Lopnur people;investigating genetic relationships between Lopnur people and other populations using bio-information resource due to provide genetic evidence for ethnological studies of Lopnur people;Methods:The mtDNA hypervariable segmentⅠand the non-coding V-region of mtDNA was measured by PCR direct sequencing method;Y-chromosome DYS287 polymorphic sites were genotyped by specific PCR method;the genetic data of Lopnur people were analyzed by softwares.Results:(1) 47 polymorphic sites were examined due to 93 mutations and 22 haplotypes;(2) mutations were common at 16223,16311,16327,16362,16298;(3) The p value of modern Lopnur people was 0.05482,variability h was 0.99604;(4)The mean number of paires differences is 6.463235±3.203367,nucloutide diversity is 0.018414±0.010212;(5) F-St and Phylogenetic tree anlysis show that modern Lopnur people presents very close relationship with Uyghur people and Caocasian;(6)The deletion frequensies of non-coding V-region was observed as 8.33%,the standard types as 91.7%;(7) Y-chromosome Alu polymorphisms were detected in moder Lopnur population.The results show that all of Lopnurian are YAP~-;Conclusion:The analyses for the polymorphism of mtDNA hypervariable segmentⅠshows that the modern Lopnur people shows a close genetic relationships with other central Asian populations,the genetic relationship between modern Lopnur people and Uyghur people in Xinjiang is the closest;The genetic data of 9bp deletion of mtDNA and DYS287 loci in Modern Lopnur people have been obtained,and they might be useful in studies of population origin,relationship between poputions,and forensic applications.
