Insulin-like growth factor I (IGF-I) is similar with Insulin in the sequence of amino acid. IGF-I takes part in many physiological functions, and it is widely used in many disease treatment of human and cell culture invitro. It is greatly required. It can't reach the requirement from microbe and cultured animal cell. In order to find a new way to get a high and stable expressing system of IGF-I from human, a gene vector of IGF-I cDNA from human lead by CaMV 35S promoter is introduced into tobacco by leaf-disc through LBA4404 (Agrobacterium tumefacients). 17 transgenic tobaccos are gained after hygromycin (20mg/L) election, GUS test and PCR identification of DNA. It shows IGF-I gene can be transcribed normal in transgenic tobacco from RT-PCR test. Furthermore, in this research a new expression vector of p1301-35S-mIGF-1-nos is constructed which include the gene of IGF-I encoded by codon preference to plant. The gene is promoted by CaMV 35S promoter which can express in tobacco in high performance. Results show IGF-I gene is introduced into tobacco and can transcribe mRNA; the vector of p1301-35S-mIGF-1-nos is fairly constructed. In this experiment, primary exploration is accomplished of the expression of IGF-I from tobacco, and it establish the fundament of IGF-I expression by tobacco later. Some information can be given out about the plant bio-reactor of IGF-I.
