Using plant bioreactor to produce pharmarciutical proteins is a novel strategy, which wasdeveloped in recent years. Compared with other bioreactor systems, such as animal bioreactor,protein prokaryotic expression system, it has the advantages like safty, proper folding andglycosylation, cost efficiency, easy operation, simple equipment. The Agro-bacteriummediated vacuum infiltration system has some additional advantages, such as time saving,high expression level, environment safety.The acidic fibroblast growth factor (aFGF) is an important member of FGF family,which can be expressed by a variety of cells from all three germ layers. aFGF shows greatpotential for clinical applications for therapy of a variety of diseases, such as Parkinson'sdisease, spinal cord contusion injury, neural regeneration in reimplantation of broken-offfinger, brain ischemia, renal ischemia, myocardial infarction, occlusive vascularitis, retinalischemia, gastric ulcer and nonhealing wound and so on.We expressed human acid fibroblast growth factor gene transiently in tobacco byAgro-bacterium mediated vacuum infiltration and studied the factors, which influenced theexpression level. The Agro-bacterium mediated vacuum infiltration system was optimizedand studied the relationship between the expression level and the cultivation time, differenttobacco materials N.b, N.H (a transgenic N.b plant which express the gene silence suppressorgene). The results indicated that the expression level was highest in the 4 days afterinfiltration and the expression level in the N.H plants was superior to that in the N.b plants.The ELISA results indicated that expression level of haFGF gene was highest in tobaccowhich was infiltrated with the pM1390-haFGF/KDEL vector, the expression leve of thewound leaf was higher than that of the intact leaf;the expression level in the materialscultivated as Valentine's method was higher than that performed by the Kapila's method.
