Bacillus subtilis fibrinolytic enzyme (BSFE) is an alkaline serine proteinase produced by Bacillus subtilis N18, which can be applied clinically as the third generation of thrombolysis medicine to prevent or to cure diseases related to thrombus, and has a promising market prospect.In this study, the gene of BSFE and its signal peptide sequence were cloned, and were subcloned into the E. coli expression vector and plant expression vector. BSFE gene and its signal peptide were introduced into wild-type Arabidopsis and tobacco by agro-bacterium mediated transformation. Transgenic plants were identified by PCR and BSFE fibrinolysis activity was assayed on the plates with swine fibrin.The results showed that the BSFE could be expressed by the host E.coli. And both the transgenic Arabidopsis and tobacco could secret target protein from leaf, stem or root. A model of expression and secretion of foreign proteins by plant tissue and root system were established, which provides some refferences for future fermentation with dustrialized production of foreign proteins.
