The most important sugar crop is sugarcane. The sucrose content of sugar was about 95% in China. There is a specific sucrose signal pathway in plant. Sucrose induce gene, for example, Sucrose synthase gene expression. Sucrose synthase is a key enzyme regulating sucrose metabolism in higher plants, which promote sucrose into a variety of metabolisms. In recent years, the main research is cloning the coding gene and biochemistry property, et al. The regulatory research of sucrose signal pathway has not been reported.In this paper, the full-length Sucrose synthase gDNA were cloned from genomic DNA of sugarcane leaf throught PCR method, named SoSs1. Using bioinformatics to analyse SoSs1 nucleotide comparability and its deduced amino acid constituent,physicochemical property and hydrophilicity/hydrophobilicity, to predict its post-translational modification,membrance spaining domain,targeting sequence,secondary structure and conserved function domain. There are some sucrose-responsive (SURE) elements in the upstream region of SoSs1 gene via bioinformatics. In the basis, we construct a couple of plant expression carriers respectively, one of which contains SURE elemet, the other has not. Then the couple of plant expression carriers are genetic transformated to Arabidopsis thaliana. Plants were sucrose-induced in vitro, northern blotting to detect the expression instance of SoSs1 gene. In addition, a SoSs1 cDNA fragment was cloned from total RNA of (Saccharum officenarum L) throught RT-PCR, named SoSuSyR. Sequence and analyze it.
