Objective: miRNA is a class of noncoding RNAs which can posttranscriptional regulate genes. miRNAs extensively take part in the growth of organism,development and disease. Currently, the difficulty on research of miRNA is the discovery of their targets. Bioinformation software predict a lot of target for miRNA,but only a handful of them have been validated by expriment. Based on the data of bioinfomationn and microarray,we try to get a set of more reliable miRNAs'targets for the further research.Methods: miRNA and mRNA profiles of HepG2 and HepG2.2.15 were compared by micoarray. Then,two database ,Targetscan and PicTar ,were used to predict the targets of miRNAs that are differently expressed in two kind of celllines. Considering that mRNA can be degraded by miRNA, the predicted targets that differently expressed between HepG2 and HepG2.2.15 were screemed by microarray. Then, these predition targets were validitied by expriments, e.g luciferase assay and miRNA over expression in cells.Result: 1. there are 23 miRNAs found to be differently expressed between HepG2 and HepG2.2.15,and we get a set of more reliable targets. 2.A recombinate adenovirus named Ad-195 was constructed which can express miR-195 in tumour cell. And It was confirmed that miR-195 regulate cell cycle by targeting CCND1 and CDK6.3.By reverse prediction, it was found that a gene named DUSP1 which is relate to stress can be co-regulated by miR-101,miR-133,miR-144,Conclusion: A set of taget genes of miRNA were found out by analysis of the data from micoarry and database.2.we found the target genes of miR-195,miR101, miR-133,miR-144,and carry out a primary research on their function.
