Ed(embryonic day)45~240 Yak upper eyelid and Ed 45~240 dorsal skin were used as test and control material respectively to investigate eyelash follicle morphogenesis. paraffin embed tissue section was stained by H.E method for studying histomorphological changes and EnVision immunohistochemistry for detecting expression of SHH andβ-catenin during eyelash follicle morphogenesis, the results is following:1. Eyelash follicle morphogenesis initiate from Ed 55 to Ed 120 , the dorsal hair follicle from Ed 65 to Ed 150 in yak;Size and growing depth of eyelash follicle bulb much bigger than dorsal that ;eyelash hair follicles are separate each other, secondary follicle is no seen ,There are secondary hair follicles and hair follicle groups in dorsal skin; Yak eyelash follicle sheath is flater and more dense than cattle's.2. SHH andβ-catenin expression during Yak eyelash follicle morphogenesis are observed in both epidermis and mesenchyme ,but SHH expression is prior toβ-catenin expression; the curve pattern ofβ-catenin expression in both epidermis and mesenchyme is consistent,β-catenin epidermis expression is more intense than the mesenchyme; the SHH expression pattern are different, It changes with stages of hair follicle morphogenesis in epidermis, and SHH mesenchyme expression is only seen in prophase and anaphase eyelash follicle.3. SHH andβ-catenin epidermis and mesenchyme expression in yak dorsal hair follicle morphogenesis occurs same trend ,but it is differ from eyelash follicle's, these two signal molecules expression intensity is weaker in dorsal hair follicle than eyelash follicle's.Our results indicate SHH andβ-catenin cooperate each other and respective function during eyelash follicle morphogenesis, The epidermis signaling dominate and the mesenchyme signaling is secondary during eyelash follicle morphogenesis in Bos Grunniens Embryos. We speculate that the time and intension difference of SHH andβ-catenin expression may play a key role in determining hair follicle types.
