OsSec27p,OsARHI are two genes of 16 up-regulated genes related vesicle trafficking response to iron deficiency in rice. Our laboratory have been blasted and analysed OsSec27p,OsARHI, and acquired OsSec27p, OsARHI full length sequence. They construct plant expression vectors CAMBIA1302-OsSec27p, pCAMBIA1302-OsARHI separately, and have been transformed into tobacco, and acquired T0 and T1 germs of OsSec27p,OsARHI, the two transgenetic tobacco.In this thesis, we have just based on the previous result in our lab, analyzing and identifying the T1 germ of transgenetic tobacco in the molecular and physical levels. Our experiment just based on the previous result from three levels, resistant select, PCR detection in gene level, and GFP detection in protein level. We used shape observation, determining the pH value and the Iron content to physical standard detection.The results of our experiment show that: the target gene fragment has been assuredly transformed into tobacco, and the gene fragment has not been deleted during the growth of tobacco. By using green-fluorescence monitor, we found that the expression of GFP is higher in the transgenetic tobacco responded to iron deficiency. Identifying the transgenetic tobaco roots and leaves, the data showed that on the condition of iron deficiency, the tolerance of transgenetic tobacco, who with OsARHI and OsSec27p, was enhance, and roots could absorb iron efficiently. Besides, the ability of iron transport in tobacco was improved. By using observation statistics and studying the growth of transgenetic tobacco, we found that in response to iron deficiency, the number of tobacco lateral root increased apparently. Furthermore, by determining the pH value of the culture, we can predict that the root of transgenetic tobacco would excrete some acid substance which would change the acidity of culture, so as to affect the abortion rate of ion in the environment.
