In the present reports, there are composed of strong promoter driving resistance gene expression in the glyphosate genetically modified crops. It made resistance glyphosate gene highly expressed in almost all plant and all developmental stages. It increase the burden on the metabolism of the plant and the plant's output is also likely to cause negative effects. In this paper, the use of glyphosate stress-induced promoter Ag2 drive the epsps gene made plant high express of resistance to glyphosate only after spraying glyphosate. In addition, the tissue-specific promoter RuBP enable epsps genes high expression in plant leaves which is the field of glyphosate pesticide spraying major part. Use the RuBP and inducible promoter Ag2 driving epsps gene at the same time should be able to enhance the anti-transgenic plants ability to glyphosate further and bring the metabolism burden of transgenic plants not too much. It can made transgenic plants with strong resistance to glyphosate and high-quality agronomic traits.So that the plant expression vector pBI121-E-M-Bt(Kanar)and pBI121-CP4E(Kanar)are based on the pBI121-E(mutations)-M-Bt (Kana) and pBI121-epsps (Kana) plasmid respectively. They constructed throμgh an intermediate vector pUC19 and the EcoRⅠsites joint transformation. Their anti-epsps gene (mutation and unmutation) common regμlated by the newly discovered inducible promoter Ag2 and the tissue-specific promoter RuBP and the selectable marker is Kana. Recombinant plasmid enters into Agrobacterium tumefaciens LBA4404 throμgh freezing and thawing method. It's transformation of Arabidopsis thaliana throμgh floral dip method and Agrobacterium-mediated. Access to the T0 generation seed, laid a material foundation for improves the resistance of plants to glyphosate by anti-epsps gene.
