1. Studies on the Differential Expression of cDNA Induced by Water Stress in Aleuritopteris argentea(Gmel.)Aleuritopteris argentea(Gmel.)is the special species in China, and it has a high drought tolerance and characteristics of resurrection plant. In order to further demonstrate the specially expression genes and the molecule mechanism in drought stress ,the spore was treated with asepsis as the material of this experiment .Some high quality mRNA was isolated from Aleuritopteris argentea by concert method which was studied by mRNA differential display. In addition, three positive differential displayed cDNA clones were obtained by modified DD techniques, such as denature PAGE, silver stain, reverse-Northern, PCR programme and no isotope. Reverse-Northern hybridization showed that one clones were up-regulated, oneclone were down-regulated one was differential expressed genes in drought stress. After sequencing, one genes were homologousb to known genes by 36%, and two genes were unknown.2. Clone of BcWRKY2 gene and promoter region in Boea crassifoliaWRKY proteins are newly identified transcription factors involved in many plant proceses including plant responses to biotic and abbiotic stresses. To date, genes ecoding WRKY proteins have been identified only from plants Boea crassifolia is the special species in China, and it has a high drought tolerance and characteristics of resurrection plant.By using designed primers in accordance with conservative sequence of WRKY , one new WRKY gene fragment BcWRKYâ…¡are obtained by RT-PCR. Subsequently full length cDNA of BcWRKYâ…¡is cloned by means of 5'RACE and sequence analysis indicates BcWRKYâ…¡belongs to group III of WRKY genes.In order to study the function of BcWRKYâ…¡deeply 5'end regulatory region sequence of BcWRKYâ…¡gene containing 908bp has been cloned from genomic DNA of Boea crassifolia by linker adaptor PCR.
