In this experiment,histological method was used to study postembryodevelopment of the ovary and uterus and effect of oxytocin and androgen on femalemandarin voles(Microtus mandarinus).The expression of estradiol and its receptorduring the estrous cycle in the ovary and uterus of the female mandarinvoles(Microtus mandarinus) by immunocytochemical method.This method was alsoused to stuy the effect of oxytocin and androgen on the expression of estradiol and itsreceptor in ovary and uterus.In order to investigate and understand the law of thereproduction in female mandarin voles and the regulate mechanism of androgen andoxytocin.1. The histological development of mandarin vole's ovary and uterus: Sort out 7groups of mandarin voles,each including 6 ones,aging from 5 day, 10 day, 15 day,20 day, 30 day, 45 day, 60 day.They were observed under light microscope.The resultsshowed that:during the ovarian development,the quantities of ovarian follicle aremore,and then ,it became little.While the types of follicles became abundant. By 5 dafter birth, there were a large number of primordial follicle. By 10 d, ovarian folliclemembrane and interstitial gland were present. By 15 d, it appeared zona pelluzida. By20 d, secondary follicles were present. By 30 d,we can see cumulus oophorusdistinctly. By 45 d, mature follicles and corpora lutea were present at small number.By 60 d ,the ovaries development finishied. The results indicated that mandarin volesreach the age of puberty on the 45th day, mature at about 60th day. Uterus keep thesame pace with development of ovary.2. On the histology research of ovary and uterus of mandarin voles during theestrous cycle: In diestrus ,there are many secondary follicles in the ovary, but italmost has no large and middle scale antral follicle.The endometrium is thiner and theuterine cavity is smaller. The estrus,inside of the ovary has many antral follicle andcorpus luteum. Several graafian follicles carry over the ovary surface. The uterinecavity expands and the uterine gland cavity expansion,internal membrane get thickest.The post partum estrus, there are 2~3 graafian follicles and many large-scale corpusluteum in the ovary.The uterine cavity diameter comparatively expands, the uterinegland cavity get smaller and the endometrium get thicker.3. The expression of the estradiol and ERβin ovary during the estrous cycle: The result of this experimental indicates that the estradiol change along with theestrous cycle in the mandarin vole's ovary obviously, and the expression of theestradiol is highest in estrus, and it is lowest in diestrus.The expression of the estradiolbetween in the estrus and diestrus has greatly significant difference. The espression ofER in mandarin vole's ovary is also presents the periodic variation.In diestrus, theexpression get weaker, but the expression get stronger in estrus.We presumed that, thefollicle growth quicker ,and the estrin secretion gradually increases adds to the mosthigh level, in estrus. But in diestrus,the follicle growth slower, and the estrin level islower, therefore creates the change of the expression on ER beta4. The expression of the estradiol and ERβin uterus during the estrous cycle:The result of this experimental indicates that in the mandarin vole's uterus theestradiol change along with the estrous cycle obviously, and the expression of theestradiol is highest in estrus, and it is lowest in diestrus. The expression of the estradiolbetween in the estrus and diestrus has greatly significant difference. The espression ofER is also presents the periodic variation in mandarin vole's uterus.In diestrus, theexpression get weaker, but the expression get stronger in estrus.5. The comparisons on post partum estrus with estrus in the mandarin voles: Theexpression of estradiol and ER during the estrous cycle and post partum estrus in theovary and uterus of the female mandarin voles by immunocytochemical method. Theresult showed the expression of estradiol and the ER all not have Significantdifference. It indicated that in the post partum estrus the estradiol and the ERβlevelare as same as in the estrus.6. The influence of oxytocin on female mandarin voles: after postnatal OTinjection,it is found that there were some cystiform follicle and many big corpusluteum, in the ovary. Endometrium thicked significantly, also the uterine gland cavityis smaller significantly. The result showed the expression of estradiol and ERβinovary and uterus is reduces significantly by immunocytochemical method.Itexplained: OT can influence the ovary and uterus in mandarin voles.7. The influence of androgen on female mandarin voles :after postnatal TPinjection,it is found that there were some cystiform follicle and the mature follicledecreased in the ovary. The endometrium thicked significantly, also the uterine glandcavity is smaller significantly. The result showed the expression of estradiol and ERβin ovary and uterus is reduces significantly by immunocytochemical method.Itexplained: androgen can influence the ovary and uterus in mandarin voles.
