DNA computer is a new type computing machine. The foundation of DNA computing is the controllable molecule operation on DNA. Some tool enzymes such as restriction endonucleases are needed for the operation on DNA molecule. The kinetics rule of the DNA computing reaction system should be mastered in order to optimize the DNA computing system and to control the reaction well. Researchers have made great effort to study this field.Firstly, this dissertation carries out the principle, characteristic and development of DNA computer; the knowledge of enzyme and enzymology; the foundation of the kinetics of reactions catalysed by enzyme. The FokI restriction endonuclease is a very important component in DNA computing system.Secondly, the research in the reaction mechanism of FokI is summarized, and two rational mechanisms are concluded. Then the math model based on the kinetics is set up.Thirdly, we introduce the application of microfluidic chip in DNA computer. We try to validate our math model through an experiment based on the microfluidic electrphoresis technology. It's a pity that we failed to do it. However, we got much valued experience.The put forward of the kinetics model of the FokI cleavage reaction is the innovation of this dissertation. Through our model, the concentrations of every component in the reaction system at some time point is known and the quantitative analysis is possible.
