Salt cress (Thellungiella halophila) is closely related to Arabidopsis and has good genetic features such as similar morphology, small genome size, short life cycle, high seed number and an efficient transformation. But, salt cress is a typical halophyte and is able to withstand dramatic salinity shock up to 500 mM NaCl. This plant does not produce salt glands or other complex morphological alterations either before or after salt adaption. Besides, salt cress and Arabidopsis share 90% and 95% identities on cDNA and amino acid sequences respectively, so it is convenient to transfer informations of Arabidopsis (gene database, protein database and mutant lines) to molecular analysis on salt tolerance of salt cress. Up to now, analysis on salt cress physiology , biochemistry and gene expression, and the construction of different cDNA libraries and mutant libraries have made salt cress a valuable model for the study of salt tolerance.This research includes: establishing the regeneration system of Thellungiella halophila and preliminarily establishing transformation system of Thellungiella halophila by transforming bar gene. We have established the regeneration system of Thellungiella halophila, then transforming bar gene mediated by Agrabacterium tumefaciens. We have studied many factors which affect transformation frequency and preliminarily established transforming system by the tag of bar gene after co-cultivation. The main results were as follows:1 The regeneration system of Thellungiella halophila has been established. In this research, the leaf explant has the highest regenerative capacity ,firstly on MS medium containing1mg/L BA+0.5mg/LNAA, then on MS medium containing MS十1mg/L BA+0.5mg/LNAA. The plant has the fine rooting capacity on 1/2MS medium.2 The transformation system has been preliminarily established. We use the leaves of Thellungiella halophila.
