With the abundance resource of wild soybean of Jilin academe of agriculture science, the experiment aim to obtain the drought-tolerance genes, supplied for gene engineering seeding.Total RNA from the seeding treated 14 hours with 20% PEG-6000 (Tester) and without treated (Driver), were extracted from the two groups of wild soybean. The difference of mRNA transcription was detected using suppressed subtractive hybridization (SSH). 362 differentially expressed cDNA fragments were cloned for composing cDNA library and identified by reverse Northern Bloting. Compared to the control, 22 of ESTs expressed under 20% PEG treated in wild soybean were sequenced. Three of them have the same sequence with another three, so 19 ESTs were analyzed in public database of GenBank.Results of BLAST analysis in GenBank showed that the drought tolerance content many processes of plant biochemistry and physiology which are controlled by many different genes in plant. Among the 19 ESTs, The ESTs Soy9 and Soy10 have 75% and 82% homology to the gene cat1 for the protein CAT, and Soy19 has 78% homology to the gene cat3 also for CAT; Soy6 has 98% homology to the gene for P5sc, Soy3, Soy7 were certified encoding gene ostA for TPS and Soy2 encoding membrane protein would be MIP, has 87% homology to the encoding gene. There are three ESTs:Soy1, Soy11 and Soy16 have not found matching sequence in the EST database, that maybe some new sequences relevant to drought tolerance that have not been studied.
