The TaVER2 is a vernalization-relative gene was screened out from the wheat( Triticum aestivum. L. cv Jingdong No.1). TaVER2 is a novel jacalin. Immumo-cytochemistory localization experiments demonstrated that vernalization could induce nuclear target of TaVER2. Posttranscriptional or posttranslational regulation of TaVER2 may play very important roles in plant development. Antisense inhibition of TaVER2 in transgenic wheat showed that maturation time were delayed compared with non-transformed wheat, suggesting that TaVER2 plays an important role in vernalization signaling and development in winter wheat.Our experiments found that over-expression of TaVER2 induces the phenotype of late-flower in the Arabidopsis(C24), the architecture of leaf was also changed in the transgenic Arabidopsis, the TaVER2 transcript level was very closely correlated with the timing of flowering in these transgenic plants indicated that the phenotype of transgenic plants was induced by the expression of TaVER2. RT-PCR experiment showed that the expression levels of FVE,FRI, VRN2 were equal while FLC was increased and LFY was decreased compared with the levels in wild type plants. Western blot analysis showed that the histone modification of H3K4~m and H3K9~m were decreased compared with the levels in wild type plants. We next examined histone modification of H3K4~m and H3K9~m at FLC by chromatin immunoprecipitation (CHIP), Compared with wild-type plants, transgenic plants showed a marked reduction in H3K4~m and a increased level of H3K9~m in regions C( the region integrets the first intron and the first exon). This result demonstrated that TaVER2 can affect the modification of histone, So inducing the change of transcript level of FLC and the phenotype late-flower of transgenic plant.Because TaVER2 is a vernalization-relative gene, I thought that vernalization was important to the transgenic plant. Our experiment showed that the phenotype of transgenic plant was early-flower after vernalization, RT-PCR and western experiment showed that modification of histone especially H3K9~m plays an important role in the regulation of plant development of transgenic plant after vernalization,...
