| Tephritidae fruit fly is one of the largest families of Diptera. There are about 4300 species in more than 450 genera through out the world. About 70 species of this family, including the Mediterranean fruit fly, Ceratitis capitata, are of the quarantine importance. Identification of fruit flies is one of critical questions in international commercial fruits and vegetables trade. It is imperative to establish rapid and reliable methods to identify and quarantine fruit flies especially in immature stages in plant quarantine. The main results were:I. In this study, 9 species (Bactrocera cucurbitae, B. tau , B. rubiginus, B. dorsalis, B. scutellata, B. parater ,B. cilifer, B. oleae , C. capitata) were selected from 4 subgenera of Bactrocera and Ceratitis genera, which are of quarantine importance. The feasibility and experiment procedure of microsatellite markers applying to quarantine fruitfly were explored. Twenty-six microsatellite loci Screened from 62 microsatellite loci could be used to differentiate the 9 species fruit fly. Break through former study thinking, adopt 0/1 situation of microsatellite loci to identify fruit fly. Just 4 PCR or 6 pairs of microsatellite primers 1 PCR could distinguish the ones. This accelerates not only the pace of quarantining in the port but also the pace of microsatellite technology applying to quarantining insect.II. Partial sequences are reported for the mitochondrial genes for cytochrome oxidase subunits I for 86 individuals of B.dorsalis. The sequences obtained are 724bp in average length and contain 33%G+C and 67%A+T and represent the character of AT biased that according to that of other insects.
