The Tiger Frog (Hoplobatrachus rugulosus) is widely distributed in South China. Because of the habitat fragmentation and destroying, also excessive human hunting, the population and distribution of the species has shrinking sharply. In this study, we choose 7 species-specific microsatellites (SSR) primers which published in GenBank and published by our lab, to study the genetic diversity and structure in three groups of H. rugulosus (populations of Fujian, Anhui and Jiangxi province).The results of the study are list as below.1. Seven micro satellite loci all showed high level of genetic diversity. In the 106 samples examined,55 alleles were detected.The number of alleles are from 6 to 10 per locus.The average was 7.86. Polymorphic information content of each locus was higher than 0.5,with the average of PIC was 0.825. The observed heterozygosity and expected heterozygosity was from 0.637 to 0.840 0.811 to 0.872, respectively. The average observed heterozygosity and mean expected heterozygosity were 0.739 and 0.849.2.Test al the loci with Hardy- Weinberg equilibrium and linkage disequilibrium, the results showed 61.9% of the loci showed significant or highly significant deviation from Hardy-Weinberg equilibrium (P<0.01 or P<0.05), while all of the loci did not show significant linkage disequilibrium between each pairs. P> 0.05.3. All three populations experienced bottleneck under IAM,TPM and SMM mutation models Using Bottleneck software.4.Use the fixed index (Fst) and Fisher's exact test to determined the level of genetic differentiation among these populations,the result shows that all the microsatellite loci did not show significant genetic differentiation among the populations, P values were higher than 0.001.5.According to Nei's unbiased genetic distance,the UPGMA tree had been constructed, which showed the genetic distance between Anhui and Jiangxi population was the smallest as 0.1146, Anhui and Fujian was largest as 0.1437.6.The gene flow has showed:Nm values was higher than 1, that is the gene flow among populations is higher. And the Fis>0 in most populations, indicating that individuals within each population had widespread inbreeding phenomenon...
