Astaxanthin, had biological activity such as antioxidation, antitumous effect, etc. The Haematococcus Pluvialis was one of the sources of natural Astaxanthin. The rapid culture method and the optimization of stress condition of Haematococcus Pluvialis was studied. The high performance liquid chromatography (HPLC) was used to analyze the extracts from Haematococcus Pluvialis and the effect of astaxanthin onheat tolerance of nematodes was measured too.The article analyzed the effect of Naphthylacetic acid (NAA),6-Benzylaminopurine (6-BA) and Kinetin(KT) of Alga Haematococcus pluvialis growth. Adding different concentrations of hormones in culture medium and testing cell density regularly. The result of single factor experiment showed that 0.5mg/L NAA、1.Omg/L 6-BA or 0. lmg/L KT can help the increase of cell density of Haematococcus Pluvialis. The optimal level combination through the orthogonal experiment was 0.5mg/L NAA、 1.Omg/L 6-BA and 0.1 mg/L KT.The effect of sodium nitrate, salicylic acid and ethephon on the accumulation of astaxanthin was evaluated. Haematococcus pluvialis was stationarily cultured in SE media with five different concentrations of sodium nitrate, salicylic acid and ethephon independently. The astaxanthin content was determined every 7days.The result showed that the astaxanthin reach rapid accumulation period in Haematococcus pluvialis cultured with sodium nitrate concentration of 0.125 g/L 7 days earlier than the controls. There were no obvious differences between the production of astaxanthin in Haematococcus pluvialis cultured in a certain concentration range (0.125~0.5 g/L) of sodium nitrate. Another result demonstrated that the astaxanthin accumulation was promoted remarkably in Haematococcus pluvialis cultured with 12 mg/L salicylic acid, the astaxanthin content in Haematococcus pluvialis reached 24.73 mg/L and 95.65% higher than the controls after 28 days culture. Therefore,0.125 g/L sodium nitrate or 12 mg/L of salicylic acid can be applied in Haematococcus pluvialis culture to produce Astaxanthin.Using high performance liquid chromatography (HPLC) method to analyze crude extract of Haematococcus pluvialis and determine the astaxanthin exists. The Astaxanthin concentration of sample was 1.47 mg/L.Using Caenorhabditis elegans as model organism, the impact of astaxanthin on resistance to stress of nematodes were investigated. Compared with the control group, the average lifespan of Caenorhabditis elegans treated with 25mg/L astaxanthin was 21.95h, which increased by 16.63%. The result showed that suitable concentration of astaxanthin can improve the heat resistance and stress resistance ability of nematode.
