| The bacteria and mycoplasma's contamination is a difficult problem in cell culture, especially the mycoplasma's. The contaminated cell will be throw away commonly, but important cell must be rescue with eliminating contamination. For so many problems to eliminate the contamination in cell culture, the studies make the BHK-21 cell, Baytril antibiotic, E.coli and M.hyorhinis as research object. With 1/10 several grades diluted measure, the Baytril's minimal inhibitory concentration was tested through the 96 well cell culture cluster for BHK-21 cell, and it is 60-80PPM. For the same method, the Baytril's MIC was tested for E.coli too, and it is 1PPM. Baytril's MIC is 0.5-2PPM for M.hyorhini. So I selected the 10PPM Baytril concentrations for the later experiment, because it can restrain the E.coli without being harm to cell.At the experiment of bacteria's section, the BHK-21cell that was contaminated artificially by E.coli, was disposed by 1×MEM with 10PPM Baytril through rinse,flowing washing,flowing washing main area,scraping,digesting,flowing washing after adherence,circular disposing,and so on. Under the Baytril's protection, the E.coli's was diluted high degree. The inspection for E.coli was done through culture medium without antibiotic at the same time. The inspection result display that the contamination was eliminated after four circulations.At the experiment of mycoplasma's section, by the same Baytril's concentration, it was almost the same method as the E.coli section to the BHK-21 cell that was infected artificially by M.hyorhinis. The inspection for M.hyorhinis was doing through DNA fluorescence dyeing and PCR at the same time. The result display that the contamination was eliminated after four circulations too, and the technique of PCR was more sensitive than the DNA fluorescence dyeing.In the end, through pairing datum sign test of biostatistics, this method has very significant effect for eliminating the E.coli and M.hyorhinis's contamination from BHK-21 cell.
