Callistemon R.Br came from the Australian and was the evergreen tree or shrub.It was the extremely exquisite omamental plant, With its unique colored posture and the widespread compatibility, It has used in the city vegetation, so it had broad city scene and the huge benefit potential. Callistemon R.Br has introduced to our country for hundred years before but not to form the scale. Simultaneously the report of the multiple differentiation heredity background of Callistemon R.Br has hardly been seen.Therefore, before the comprehensive utilization research, it has the extremely important theory and practical significance that the existing every germplasm resources were appraised and clarified the blood relationship and the Molecular Systematics among them.The genetic diversity of fifteen Callistemon resourses were studied with the methods of morphological marks and ISSR molecular marks for the first time.The results were as the following:The botanical and biological characters of fifteen Callistemon resourses were measured with morphological marks, nine germplasm resources with values were sifted. The data was analysised by the software SAS6.2 and the data has achieved the extremely remarkable level. The germplasm resources were classified with the method of normalization of standard derivation.Callistemon R.Br was divided two kinds When the clustering coefficient was 45.697. the first big kind included C. citrinus, C. comboynensis, C. formosus, C. injune pink , C. linearis , C. paludosus , C. polandii, C. rigidus , C. salignus , C. salignus rubra , C. teretifolius , C. violacens , and the second kind included C. brachyandrus, C. pachyphyllus green, C. viminalis.The modified CTAB method was used to extract genomic DNA with good results.The optimalized ISSR-PCR condition sresults:1×PCRbuffer,2.5mmol.L-1Mg2+,0.2mmol.L-1dNTP, 0.4μmol.L-1 primer, 1.2ng.μL-1template DNA, 0.5U Taq DNA poly merase. Reaction progran: pre-denaturtion at 94℃ for 3 minutes,then denaturtion at 94℃ for 0.5 minutes,annealtion at 50℃ for 1 minute,extension at 72℃ for 1.5 minutes,45 cycles in all.Using the optimal experimental system, the ISSR member distinction and the blood relationship relationship analysis of the one Melaleuca and 15 Callistemon R.Br kinds have carried on for the first time.The results indicated that, a totle of 220 unambiguous bands were got with 19 primers, out of them 212(96.4%) were polymorphic.The Dice genetic coefficient ranged from 0.451-0.732, thus...
