Objective: Implantation is initiated, when direct interaction between embryo and the uterus occurs, by the recognition of hatched, and free embryo with the surface of uterus. Successful implantation results from the fine regulated co-operation between the invasive blastocyst and the receptive endometrium. It has been known that the stage specific expression of sLex oligosaccharides has an important role in embryo implantation. Fucosyltransferases (FUTs) are the key enzymes for fucosylated oligosaccharides synthesis. Thus, the studies related to the regulation of FUT7 expression may provide a new idea and approach to control implantation process.Methods: Using the in vitro model of mouse uterine endometrial epithelial cells, FUT7 expression was analyzed by RT-PCR, indirect immunofluorescence and Western blot methods and to investigate the mechanism of implantation related to estrogen and FUT7 antisense oligodeoxynucleotide regulation.Results: Our results showed that estrogen up-regulated FUT7 gene and enzyme expression , while FUT7 antisense oligodeoxynucleotide down-regulated FUT7 gene and enzyme expression in mouse uterine endometrial epithelial cells. These data indicate that estrogen could stimulate the expression of FUT7, which make more synthesis of sLe~X located on the surface of uterine epithelial cells it may constitute an implantation network by estrogen→FUT7→sLe~X.Conclusion : The results suggests that estrogen may regulate...
