Implantation is a complex developmental process that is initiated by adhesion of the embryo to the endometrial epithelium. The trophoblast of blastocyst establishes contact to the surface of the uterine epithelium is critical for the successful implantation of mammalian embryos. At the maternal-fetal interface, the selectin oligosaccharide -based ligands, such as endometrial cells (sialyl lewis x) is specifically expressed on human uterine epithelial cell, and it can bound to the selectin located on the surface of the trophoblasts. The increased synthesis of endometrial cells is related to the receptivity. of the uterine endometrium. The selectin-ligand adhesive systems may be critical to the establishment of human implantation. Fucosyltransferases (FUTs) are the key enzymes for fucosylated oligosaccharide synthesis. It has been known that the stage specific expression of sLe~x oligosaccharides has an important role in embryo implantation. Thus, the studies of effect of oligosaccharides may provide a new idea and approach to control implantation process.Antisense oligodeoxynucleotide (AS-ODN) is the synthetic DNA molecule, which has complementary sequences to the mRNA of target gene. By base pairing, AS-ODN interferes the expression of the target gene by binding to mRNA, inhibiting translation, or degradating the target mRNA.To investigate the changes of the FUT7 and sLe~X oligosaccharide expression and probed into the mechanism of the role of sLe~X in implantation,using the in vitro model which consisted of human uterine endometrial epithelial cell line (RL95-2) and human trophoblast cell line (JAR) transfected by the antisense oligodeoxynucleotide of FUT7, RL95-2 cells were analyzed by RT-PCR, Western blot and indirect immuno- fluorescence methods.
